Nació el 4 de septiembre de 1898 en Carbondale, Illinois (Estados Unidos).
Participó en largometrajes cómicos en la Essanay, aunque se hizo popular en el año 1921, cuando, en El Caid (The Sheik), de George Melford, trabajó junto a Rodolfo Valentino.
Interpretó a menudo para Cecil B. De Mille, quien la dirigió en: El señorito primavera (The Affairs of Anatol, 1921) y Los diez mandamientos (1923), entre otras. En 1926 volvió a encarnar un papel junto a Valentino con El hijo del Caid (G. Fitzmaurice), aunque su carrera finalizó cuando acabó el cine mudo. Intentó regresar al cine en 1937 con Almas en el mar (Henry Hathaway),
Muerte
Agnes Ayres falleció el 25 de diciembre de 1940, a causa de una hemorragia cerebral.
