Duplass nació el 7 de diciembre de 1976 en Nueva Orleans, Louisiana, hijo de Cynthia "Cindy" (de soltera Ernst) y Lawrence "Larry" Duplass. Fue criado en una familia católica, y asistió a la Secundaria Jesuita. Es el hermano del director y escritor Jay Duplass. Está casado con su co-protagonista en The League, Katie Aselton. Tienen dos hijos. También es el cantante de la banda de rock Volcano, I'm Still Excited!!.
